Amphipathic Nature: The most tested property of membrane lipids (phospholipids) is that they contain both hydrophilic (polar) heads and hydrophobic (non-polar) tails.

1. Elemental Composition and Structure

Lipids are a diverse group of macromolecules, but they share a defining characteristic: they are chemically distinct from carbohydrates, proteins, and nucleic acids due to their elemental makeup and solubility.

Typical Composition

Lipids are composed principally of Carbon (C), Hydrogen (H), and Oxygen (O). While carbohydrates typically follow a 1:2:1 ratio (CH₂O)_n, lipids contain a much higher proportion of carbon and hydrogen relative to oxygen. This abundance of C-H bonds makes lipids excellent for long-term energy storage.

Note on Heteroatoms: While simple lipids (fats/oils) are C, H, and O, complex lipids like phospholipids also contain Phosphorus (P) and sometimes Nitrogen (N) in their polar head groups.

Fatty Acid Tails

The fundamental building block of many lipids is the fatty acid. A fatty acid consists of two distinct parts:

Carboxyl Group (-COOH): An acidic functional group at one end.
Hydrocarbon Chain: A long chain of carbon atoms bonded to hydrogen. This chain is non-polar and responsible for the lipid's hydrophobic properties.

2. Solubility Vocabulary: Interacting with Water

Understanding the behavior of lipids in biological systems requires precise vocabulary regarding how molecules interact with water.

3. Amphipathic Molecules

The term amphipathic (from Greek amphi meaning "both" and pathos meaning "feeling") refers to a molecule that possesses both hydrophilic and hydrophobic properties.

This is the single most important concept regarding biological membranes. The primary example of an amphipathic molecule is the phospholipid.

The Head (Hydrophilic): Contains a phosphate group (negatively charged) and glycerol. It interacts readily with the aqueous environment (cytoplasm and extracellular fluid).
The Tails (Hydrophobic): Consist of two long fatty acid chains. They repel water and seek to aggregate with other lipids.

4. Lipid Structures in Aqueous Solution

Because lipids are amphipathic, they spontaneously arrange themselves in water to achieve the lowest energy state. This is driven by the hydrophobic effect—water molecules exclude non-polar tails to maximize their own hydrogen bonding and entropy.

Depending on the shape and concentration of the lipids, three main structures form:

1. Micelles

Micelles are spherical structures formed by fatty acids or single-tailed phospholipids. The hydrophobic tails tuck inside the sphere, while the hydrophilic heads form the outer surface protecting the core from water. This is how soap traps grease (hydrophobic) inside the micelle to wash it away.

2. Lipid Bilayers (Liposomes)

Because membrane phospholipids have two tails, they are too bulky to form a tight micelle. Instead, they form a bilayer (sandwich). The hydrophobic tails face each other in the middle, and the hydrophilic heads face the water on both sides. This is the structural basis of the cell membrane.

Clinical Connection: Liposomes
Liposomes are artificial vesicles (hollow spheres made of a lipid bilayer). They are frequently used in medicine for drug delivery. A hydrophilic drug can be trapped in the watery center, or a hydrophobic drug can be trapped in the lipid membrane itself, allowing transport through the bloodstream to target cells.
