1. Enzymes (Catalysis)

Enzymes are arguably the most tested protein function on the DAT. They act as biological catalysts.

Key Characteristics

Specificity: Enzymes bind to specific substrates at their active site.
Reusability: Enzymes are not consumed in the reaction; they can be used repeatedly.
Efficiency: They can increase reaction rates by factors of millions.

Lock & Key vs. Induced Fit: The "Lock and Key" model suggests the enzyme and substrate fit perfectly like puzzle pieces. The "Induced Fit" model (more accurate) suggests the enzyme undergoes a slight conformational change upon binding to fit the substrate more tightly.

Mechanism of Action

Enzymes work by stabilizing the transition state, thereby lowering the activation energy needed for the reaction to proceed. Remember: They do not alter the free energy change ($\Delta G$) of the reaction.

2. Hormones (Signaling)

Protein hormones circulate in the blood to transmit signals between organs. Note that not all hormones are proteins (some are steroids/lipids).

Peptide Hormones

Solubility: Water-soluble (Hydrophilic).
Transport: Move freely in the blood.
Action: Cannot cross the cell membrane; must bind to extracellular receptors to trigger a second messenger cascade.
Examples: Insulin, Glucagon, ACTH, FSH, LH.

3. Receptors (Membrane Signaling)

Receptors are proteins embedded in the cell membrane (integral proteins) or located inside the cell (intracellular) that bind to signaling molecules (ligands).

G Protein-Coupled Receptors (GPCRs)

This is a massive family of cell surface receptors particularly important for the DAT. They span the membrane 7 times.

Ligand binds to the GPCR.
GPCR undergoes a conformational change.
This activates an associated G-protein (exchanging GDP for GTP).
The alpha subunit dissociates and triggers a cellular response (often via cAMP).

4. Immunity (Antibodies)

Antibodies (Immunoglobulins) are specialized proteins produced by B-cells (plasma cells) that identify and neutralize foreign objects like bacteria and viruses.

Structure

Y-Shape: Composed of 2 Heavy chains and 2 Light chains.
Variable Region: The tips of the "Y" containing the antigen-binding site. This region is unique to each specific antibody.
Constant Region: The stem of the "Y", determines the class of antibody (IgG, IgM, IgA, etc.).
Disulfide Bridges: Covalent bonds holding the chains together.

IgA (Found in mucosal areas, saliva, breast milk)

5. Structure (Support & Strength)

Structural proteins provide the framework for cells and tissues.

6. Storage (Amino Acid Reservoir)

Ferritin: Stores iron in the liver.

Ovalbumin: Protein in egg whites (amino acid source for developing embryo).
